Amitriptyline is
prepared from ch co, con (CH3)2 dibenzocycloheptenone. The ketone is
synthesized by condensation of phthalic anhydride with phenylacetic acid to
give enzalphthalide, which on reduction
yields 2phenethylbenzoic acid; this is converted to the acid chloride, which on
treatment with aluminium chloride undergoes cyclization. The ketone is reacted
with the appropriate Grignard reagent,
followed by hydrolysis of the complex and dehydration using acetyl chloride to
get amitriptyline.
Amitriptyline
hydrochloride (Brand Names : Amitid; Amitril; Deprex; Domical Elavill; Endep;
Lentizol; Miketorin; Saroten; Tryptizol) is an odourless, colourless or white
powder. It is soluble in water (1 in 1), alcohol (1 in 1.5) chloroform (1 in 2)
and methanol (1 in 1); practically insoluble in ether; m.p. 196-197* stored in
a well-closed container.
Uses :
Amitriptyline
is a tricyclic antidepressant used in the treatment of mental depression, and
for nocturnal enuresis in children.
Side Effect
Many side-effects
of amitriptyline and other tricyclic antidepressants are caused by their
antimuscarinic action. These include dry mouth, sour or metallic taste,
constipation, urinary retention, blurred vision and disturbances in
accommodation, palpitation, tachycardia, and impotence. Many other side effects
have also occurred.
