AMODIAQUINE

Amodiaquinone
contains a diethylamino-substituted amino-group in position 4 of quinoline and a chlorine atom in
position 7; the substituent attached at C-4 is aromatic and possesses a
phenolic group. For its preparation, 4-amino-2- (diethylaminomethyl)phenolis
synthesized from p-nitrophenol which is then condensed with fomaldehyde and
diethylamine related to the Mannich reaction, followed by removal of the
protective group by hydrolysis.
This
4-amino-2-(diethylaminomethyl) phenol condenses with 4, 7-dichloroquinoline to
yield amodiaquine.
Amodiaquine,
m.p. 208°, is a crystalline base which is used medicinally in the form of its
dihydrochloride. It is stored in airtight containers.
