It is the salt of a diamine, two moles
of penicillin are available from each molecule of the salt. It is prepared by
adding an aqueous solution of one mole N,N-dibenzylethylenediamine diacetate to
an aqueous solution of moles sodium penicillin G
Benzathine penicillin G is a white
odourless, tasteless, crystalline powder, m.p. 123-124°; very insoluble in
water (1 g in 5000-6000 ml). This solubility property gives the compound great
stability and prolonged duration of effect. It is soluble in alcohol (1 in 65);
practically insoluble in chloroform and ether At the pH of gastric juice it is
quite stable, and food intake does not interfere with its absorption. It is
stored at a temperature not exceeding 30 in airtight containers and protected
from light and moisture
Uses
1.
Benzathine penicillin has the same
antimicrobial action as benzylpenicillin, but because of the relatively low
blood concentrations of benzylpenicillin produced, its use should be restricted
to micro-organisms that are highly susceptible to benzylpenicillin.
2.
It is used for the treatment of syphilis and
other treponemal infections, such as yaws, pinta, and endemic syphilis.
3.
It may be used to cure streptococcal
infections, particularly of the upper respiratory tract. It should not be given
to patients suffering from gonorrhoea.'
