Cortisone may be prepared by partial or total
synthesis starting from ergosterol, progesterone and sapogenins
Cortisone, m.p. 220-224°, occurs as platelets. It is fairly
soluble in cold methanol, ethanol, acetone; much less soluble in ether,
benzene, chloroform; slightly soluble in water. It gives orange-red solution
with intense green
fluorescence in concentrated sulphuric acid. It is used primarily as the
21-acetate due to its increased stability.
Cortisone Acetate (Brand Names: Cortistab;
Cortelan; Cortisyl artriona; Compound E acetate; Cortogenacetate), mp 235-238,
is a white, odourless, crystalline powder. It is practically insoluble in
water; soluble in alcohol, acetone, chloroform and dioxane; slightly soluble in
ether and methyl alcohol. It reduces ammonical silver nitrate solution at room
temperature. Solution in sulphuric acid gives a yellow solution without
fluorescence. It is kept in well-closed containers and protected from light.
Uses:
It Betamethasone is a glucocorticoid
used as antiinflammatory and immunosuppressant glucocorticoid agent. Cortisone acetate has been used in the
treatment of many allergic and inflammatory disorders. It is given in the
treatment of rheumatoid arthritis including gouty arthritis, ulcerative
colitis, regional enteritis, nephrotic syndrome, scleroderma dermatomyositis,
psoriasis, periarteritis nodosa, pulmonary granulomatosis and alcoholism.
