Cyclobarbitone contains a cyclohex-1- enyl group in the
5-position. It is prepared by condensing ethyl cyanoacetate with cyclohexanone
in the presence of sodium ethoxide or pyridine to yield ethyl
cyclohex1'-enylcyano acetate. With sodium ethoxide and ethyl iodide this ester
is alkylated to give ethylcyanoethylacyclohex-1'-enylacetate. This reactive
cyano-ester is condensed with urea to form an open-chain cyanoureide.
Hydrolysis of the -CN group afforded an acid which cyclizes spontaneously to
cyclobarbitone.
It occurs as shiny crystals, insipid bitter taste, m.p. 171174;
very slightly soluble in cold water, appreciably soluble in hot water. One gram
dissolves in 5 ml alcohol, and 20 ml ether. It is stored in airtight
containers.
Uses : It has central depressant
(hypnotic, sedative) activity used as an anaesthetic and sedative.
Cyclobarbitone Calcium : (Brand Names : Hexemal calcium; Itridal,
Kollerdormfix; Pronox; Cyclobarbital calcium) is a white or slightly yellowish,
crystalline powder. It is slightly soluble in water and alcohol; practically
insoluble in chloroform and in ether; stored in airtight containers; used as
hypnotic and sedative.
