CYPROHEPTADINE
Brand Name : Periactinol.
Cyproheptadine is synthesized from a ketone consisting of two
benzene rings fused in a linear fashion to a seven membered ketone. It is first
brominated with N- bronosuccinimide (NBS) and then dehydrobrominated by
treatment with triethylamine. The product is reacted with the Grignard
reagent, N-methyl-4-piperidyl magnesium chloride, to yield a complex. This is
dehydrated by solution in acetic acid and then treated with hydrogen chloride
and acetic anhydride to yield cyproheptadine hydrochloride.
Cyproheptadine, m.p. 112-114, is a crystalline compound.
Cyproheptadine
hydrochloride (Brand Names : Anarexol; Antegan; Ifrasarl; Nuran; Periactin;
Vimicon; Cipractin; Peritol), Imp. 252-254 (decomp), is a white to slightly
yellow, odourless, crystalline powder which is soluble in water, alcohol,
methanol and chloroform; practically insoluble in ether.
Uses: Cyproheptadine is an antihistamine, antiserotonin and
antipruritic agent. It has antimuscarinic activity and is used to suppress the
symptoms of allergic reactions and is effective in cold urticaria . It is also
used as an appetite stimulant, in eatment of vascular and migraine headaches,
Cushings disease and allergic conditions and to relieve various itching skin conditions.
Side Effect
The side effects are the same as for promethazine. Increased
appetite and weight gain may occur.
