Dapsone is prepared by reduction of the corresponding
dinitro-compound. The dinitro product cannot be preprered by nitration of
diphenyl sulfone, since the -SO, group, like -SOH, is meta-directing and,
therefore, 3,3'-and not the 4,4'-dinitro-compound is obtained. Dapsone is
synthesized by interaction of 4-chloronitrobenzene with sodium sulphide to give
di-(4-nitrophenyl) sulphide, which is then oxidized by chromic acid to the
sulphone.
Dapsone is a white or slightly yellowish white odourless,
crystalline powder, m.p. 175-176°, soluble in alcohol (1:30); freely soluble in
acetone and dilute mineral acids; practically insoluble in water. -
The pure compound is light-stable, but in the presence of traces
of impurities, including water, it becomes photosensitive and it is
discolourized. It forms soluble salts with acids, and gives the ordinary
reactions of a primary aromatic amine. The drug is protected from light.
Uses
Dapsone is antibacterial (leprostatic), and dermatitis herpethermis
suppressant. Therefore, it is used to treat both lepromatous and tuberculoid
types of leprosy. It is cheap, relatively nontoxic and highly active against
Mycobacterium leprae
