DEXTROPROPOXYPHENE
Brand Names : d-Propoxyphene
This aliphatic tertiary amine base is also an arylsubstituted
aliphatic ester of propionic acid. The substituted isobutyro-phenone required
for its synthesis can be prepared by condensation of propiophenone,
paraformaldehyde and diethylamine hydrochloride (Mannich reaction). The
aminoketone so obtained reacts with the Grignard reagent, benzyl magnesium
chloride, which on hydrolysis gives mainly the form of a tertiary alcohol
hydrochloride. This alcohol is esterified by refluxing with propionic anhydride
in the presence of pyridine.
Dextropropoxyphene. m.p. 75-76°, occurs as crystals.
