Diloxanide furoate is an aromatic ester of 2-furoic acid. For its
synthesis, 4-aminophenol is treated with chloral cyanohydrin in the presence of
pyridine and triethylamine. The resulting phenol is then benzoylated before
methylation of the amido group, followed by hydrolysis
with 0.5 N sodium hydroxide to give diloxamide. Esterification
of diloxanide with furoyl chloride yields diloxanide furoate.
Diloxanide furoate is a white, odourless, crystalline powder; very
slightly soluble in water; slightly soluble in ether and alcohol. It is
protected from light .
Uses
It is a luminal amoebicide used in the
treatment of intestinal amoebiasis. With metronidazole it is used to treat
Entamoeba polecki infection.
Adverse effect
Flatulence is the most common adverse effect during treatment with
diloxanide furoate. Vomiting, pruritis and urticaria may occasionally occur.
