Doxycycline, m.p. 201° (decomp.), was first obtained  by a chemical 
transformation of oxytetracycline but it is now synthesized by catalytic reduction of methacycline or by reduction of a benzyl mercaptain derivative with Raney nickel catalyst. In the 6-α-methyl epimer, the orientation of 6-methyl group is different from that in the tetracyclines. The 6a-methylepimer is three times more active than its 6α-epimer. The absence of the 6-hydroxyl group produces a compound that is highly stable to acids and bases and that has long biologic half-life. It is also very well absorbed from the gastrointestinal tract, thus allowing a smaller dose to be administered.

HOMATROPINE

HOMATROPINE Brand Names : Mandelyltropeine, Tropine mandelate. Homatropine is a synthetic alkaloid which is a lower homologue of atropine. It is prepared by the interaction of tropine and mandelic acid. Homatropine, m.p. 99-100°, occurs as colourless, odourless crystals or a white crystalline powder. It is hygroscopic, but only slightly soluble in water; soluble in alcohol, benzene, chloroform, ether, acetone and dilute acids. The hydrobromide or hydrochloride is obained readily from the free base by neutralizing with hydrobromic or hydrochkoric acid. The hydrobromide occurs as colourless, odourless crystals or a white crystalline powder. It is soluble in water and alcohol; slightly soluble in chloroform; very slightly soluble in ether; stored in airtight containers and protected from light.
