Blue.
Evans blue is an azo-dye ofdiagnostic value and is prepared by
coupling tetrazotized o-tolidine with 2 moles of Chicago acid
(8-amino-1-hydroxy-5,7-naphthalenedisulphonic acid) in alkaline solution.
Evans blue is green, bluish-green or brown, odourless powder. It
is very soluble in water; very slightly soluble in alcohol; practically
insoluble in carbon tetrachkoride, chloroform and ether; at pH 10 the colour is
changed. It is destroyed by strong oxidizing and reducing agents and
precipitated from saturated solution of neutral salts. It is rather stable in
aqueous solution. and may be autoclaved at l5 lbs pressure for 30 minutes. The
aqueous solutions are quite stable and may be autoclaved. Saline solutions are
less stable and should not be autoclaved.
In body Evans dye combines firmly with the plasma albumin. The
colour developed is directly proportional to its concentration.
Uses : Evans dye is used for the determination of blood (or
plasma) volume. A quantitative amount of the dye is injected intravenously.
After a suitable interval a sample of blood is taken, and the dye in the plasma
is determined colorimetrically. The dye does not escape into the extracellular
fluid and is only slowly excreted via the kidneys.
