Glyceryl trinitrate is the glyceride of nitric acid. It is
prepared by slowly adding glycerol to an ice-cold mixture of concentrated
nitric acid and concentrated sulphuric acid in the ratio of 1:4. The solution
is carefully passed into ice-cold water when the glyceryl trinitrate is
separated as a heavy oil. It is purified by repeated washing with cold water.
Glyceryl trinitrate is a heavy, pale yellow, oily, inflammable,
explosive liquid with sweet burning taste. It produces headache on tasting and
explodes on rapid heating or when struck with hammer. In impure form it may
explode spontaneously. It is slightly soluble in water; soluble in methyl
alcohol, alcohol, carbon disulphide, acetone, chloroform, ether and in glacial
acetic-acid.
Concentrated
glyceryl trinitrate is a 9-11% solution of glyceryl trinitrate in alcohol
(96%). Dilute nitroglycerin is a mixture of glyceryl trinitrate with lactose,
glucose, alcohol, propylene glycol or other suitable inert excipient.
Uses
Glycery trinitrate relaxes smooth
muscles, including vascular muscles and reduces the blood
pressure. It is used as a vasodilator in the prophylaxis and treatment of
angina pectoris; for bronchial asthma în dogs.
Adverse Effect
Adverse effects of glyceryl trinitrate
are acute poisoning, occurring in industrial workers; nausea, vomiting,
abdominal cramps, headache, mental confusion, delirium, bradypnea, bradycardia,
paralysis, convulsions, methemoglobinemia, cyanosis,
circulatory collapse and death.
