Isoniazid, the hydrazide of isonicotinic acid, is synthesized from
Y-picoline, which by permanganate oxidation is converted in the normal manner
into isonicotinic acid. The acid is esterified by means of " ethanol and
concentrated sulphuric acid. Direct N2 condensation of the ester with hydrazine
affords the required hydrazide.
lsoniazid occurs as colourless, odourless crystals, or white
crystalline powder, m.p. 171.4°. soluble in water (1 : 8) and alcohol
(1 : 50); slightly soluble in chloroform and ether. The drug is stored in
well-closed containers and protected from light
.
Uses
Isonlazid is antibacterial
(tuberculostatic), i.e., it has N bacteriostatic effect on the tubercle bacilli
therein. It is used to treat pulmonary and extrapulmonary tuberculosis.
Pyridoxine N is often given in conjugation with isoniazid to avert its
neurotoxicity .
