Mercaptopurine, m.p. 313-3149 (decomp),
is a yellow, odourless, crystalline powder. It is practically insoluble in
water, acetone and ether; slightly soluble in alcohol; soluble in hot alcohol
and in solutions of alkali hydroxides; slightly soluble in M sulphuric acid. It
is stored in air-tight containers and protected from light.
Mercaptopurine acts as an antimetabolite
for adenine in the synthesis of nucleotides by living cells. It is thought to
act by entering leukaemic blood cells and interfering with their function.
It
is metabolized in the body to 6-thiouric acid. After the intracellular
conversion of mercaptopurine to active nucleotides, it interferes with nucleic
acid synthesis.
Uses: Mercaptopurine is an antineoplastic
agent which acts as an antimetabolite. It also has immunosuppressant
properties. It is used, usually with other agents, in the treatment of
leukaemia and various auto-immune disorders.
Side Effect
The side-effects of mercaptopurine include bone-marrow depression,
hypoplasia and hepatotoxicity.
