Methacycline is synthesized by chemical modification of
oxytetracycline. Its antibiotic spetrum is similar to the other
tetracyclines but its potency is greater than the other such compounds. The
greater stability of methacycline is due to the modification at carbon atom 6.
The substitution of the 6-hydroxy group markedly increases the stability of
ring C to both acids and alkalies, by preventing the formation of
anhydrotetracyclines by acids and ofisotetracyclines by bases.
Methacycline hydrochloride, m.p. 205 (decomp.), is a yellow to
dark yellow crystalline powder.
It is slightly soluble in water (1 in 100) and
in alcohol (1 in 300); practically insoluble in chloroform and ether; soluble
in dilute solutions of Sodium hydroxide. It should be stored in tight and
light-resistant containers in a cool place
