Methadone is prepared by condensing diphenylmethyl cyanide with 2-chloro-1-dimethyl aminopropane in the presence of sodamide. The intermediate cyclic
quaternary ammonium salt undergoes cleavage to yield a mixture of amines. The
amines are hydrolyzed to a mixture of ketones which are separated. One of these
ketone is methadone.
Methadone hydrochloride, m.p. 235°, is a colourless, odourless
crystalline powder with a bitter taste. It is soluble in water; freely soluble
in alcohol and chloroform; practically insoluble in ether and glycerol. A 1%
solution in water has a pH of 4.5 to 6.5.
Uses: Methadone is a potent
analgesic with actions, uses and side effects similar to those of morphine. It
is used to alleviate many types of pains and has a depressant action on cough
Centre.
The toxicity of methadone is 3 to
10 times greater than that of morphine. It has analgesic effect twice that of
morphine and relatively greater respiratory depressant effect than morphine.
Pulmonary oedema after over dose is common.
