Metronidazole is an imidazole or cyclic amidines which can be
synthesized by condensation of compounds containing adjacent carbonyl group
(i.e.-CO-CO) with ammonia and formaldehyde (Radziszewski's reaction).
Tartaric
acid dinitrate and acetaldehyde yields, after thermal decarboxylation,
2-methylimidazole. Nitration, followed by alkylation in the 1-position with
ethylene chlorohydrin yields metronidazole.
Metronidazole is a pale yellow or cream-coloured crystalline
compound, m.p. 150-160; has limited solubility in water and other organic
solvents. It is stable in air but darkens on exposure to light. Therefore, it
is protected from light
Uses
Metronidazole is an antiprotozoal substance and used in bacterial
infections, anaerobic of malignant neoplasms. Darkened urine may occur when the
drug is given in higher doses. The yellow pigment may be a metabolite of
metronidazole. The adverse effects of the drug are gastro-intestinal
disturbances, nausea, unpleasant taste, headache, anorexia and vomiting.
