Promethazine is a phenothiazine derivative which is prepared by
condensing phenothiazine with CICH2 CH(CH3)N (C2H5)2 in the presence of
sodamide.
Promethazine is prepared from 10-phenothiazinepropyl chloride and
dimethylamine in the presence of copper: and from Grignard complexes of
dimethylaminopropyl halide and phenothiazine.
Promethazine, m.p. 60°, is a crystalline compound and used in the
form of hydrochloride salt. -

It is a white, odourless powder which is very soluble in water; soluble
in alcohol and chloroform; practically insoluble in ether, acetone and ethyl
acetate. It turns blue on prolonged exposure to air and moisture. It is
keptinairtight containers and protected from light.
Uses : Promethazine is an antihistaminic agent having some
antimuscarinic, antiemetic, antiserotoninergic and local anaesthetic
properties. It is given for allergic conditions and to treat burns.
Side Effect
The side effects include sedation, drowsiness, lassitude,
dizziness, and inco-ordination. Cardiovascular side-effects are more commonly
observed after injection. Bradycardia, tachycardia, increase in blood pressure,
hypotension, jaundice and thrombocytopenic purpura have been reported.
