Sulphobromophthalein sodium is a phthalein derivative used as a
dye in tests of liver function. It is prepared by dissolving phthalic
anhydride in fuming sulphuric acid, followed by bromination with iodine as a
catalyst. The tetrabromophthalic anhydride is then condensed with phenol and
the resulting tetrabromophenolphthalein is sulphonated and converted to
the disodium salt.
It occurs as a white, bitter, odourless, crystalline, hygroscopic
powder; soluble in water; practically insoluble in alcohol and in acetone. The
presence of sulphonic acid groups into the compound decreases the toxicity and
increases the water solubility. The compound is injected intravenously and the
amount remaining in the blood after a certain time interval is determined
colourimetrically. The rate at which the dye is removed from the blood is a
measure of the hepatic function.
Uses: Sulphobromophthalein sodium is used as a diagnostic agent
for testing the functional capacity of the liver. The test is usually performed
in the morning after the patient has taken a fat-free breakfast. No food must
be given during the test. The concentration of the dye in the bloodstream is
measured at the end of 1 hour and at regular time intervals thereafter to
determine the rate of clearance.
Side Effect
Allergic-type reactions have been reported in patients given
sulphobromphthalein. Inadvertent subcutaneous infiltration may cause irritation
and necrosis.
